The as-yet-untitled British production, which also stars Girish Karnad and Om Puri, is a family drama focusing on a father and his three daughters.
Speaking to Bollywood Hungama, Khan said: "Yes, mum and I are doing a film together. It's an absolutely lovely script and shooting starts in May in London."
The movie is expected to be showcased at the London Film Festival in October.
Tagore previously featured alongside her son Saif Ali Khan in Vidhu Vinod Chopra's Eklavya: The Royal Guard in 2007.
