DreamWorks will make five or six films a year under the terms of the deal, which accounts for over a third of its total $825 million production budget.
In return, Ambani's Reliance BIG Entertainment will control marketing and distribution of the studio's films in India. The Walt Disney Company will be responsible for distribution in the rest of the world.
Spielberg hailed the arrangement as a "visionary step", saying that it gives his firm "a new set of dreams to work towards".
DreamWorks ended its previous partnership with Paramount Pictures last year. The collaboration had been behind box office hits including Transformers and Blades of Glory.
