The Bollywood star has asked for Kangna Ranaut and Zarine Khan to be considered for the lead female roles instead, according to Mid Day.
Salman is taking over the lead from Anil Kapoor, who starred in the first film No Entry.
Director Anees Bazmi told the tabloid: "Men cheating on their wives... both Salman and I feel we can't get away from that theme. It's like the Superman franchise.
"When audiences go to see a Superman film they expect to see Superman flying."
On relegating Kapoor to a minor role, he added: "We need to be realistic. If Salman is in a film all other actors take a backseat."
The director also admitted that Salman is deeply involved in the scripting of the film, which is his second with Bazmi after the hit Dabanng.
It was claimed that one of the reasons Basu broke up with long-term boyfriend John Abraham was that the actress was trying to get a starring role opposite Salman.
