The Udaan director reportedly grew impatient waiting for Khan to wrap up his other commitments and has decided to go ahead without the box office name, Mid Day reports.
Kashyap told the paper: "I am going ahead with Bombay Velvet without Aamir. He is a very busy man and I have waited long enough. I don't even know if he wants to do the film."
Bombay Velvet is an adaptation of historical work Mumbai Fables, which is set in post-independence India in the 1960s.
The book, by Gyan Prakash, aims to tell the history of the old city through the many intersecting stories of its residents.
There are rumoured to be concerns that the budget for the film will be much lower than before because Khan is no longer in the running for the lead role.
However, Kashyap said: "Studio 18 is producing the film and money is not an issue here."
John Abraham, Naseeruddin Shah and Kay Kay Menon have also joined the cast of the movie, according to Mid Day.
It will begin shooting after Kashyap finishes his current projects That Girl in Yellow Boots and Gangs of Wasseypur.
Khan recently revealed that he will be playing a bigger role in the upcoming sequel to the hit Delhi Belly, which will be titled Disco Fighter.
