The actress appears in the 2006 film, which was itself a remake of the 1978 movie starring Amitabh Bachchan.
Koppikar played Shah Rukh Khan's girlfriend Anita in the remake, which follows the accomplice of a crime lord who finds himself being chased by the police.
She told New Kerala: "I'm not a part of Don 2 and I don't regret not being a part of it."
Koppikar, 34, claimed that she is more excited about her delayed film Shabri.
Shabri, which was produced by controversial director Ram Gopal Varma, has taken five years to arrive in cinemas.
The movie centres on a woman who finds herself caught between the mafia and the police when circumstances get out of control.
Koppikar said: "This film was made a couple of years back and now it is releasing. Better late than never. I'm very excited.
"Everyone has worked very hard on it and I'm glad it's releasing. The promotions have just started."
Don 2 arrives in cinemas on December 23.
