Dhawan said that the actors are a "dream team" to work with.
"Both Akshay and John are extremely disciplined and professional. They were always on time and focused through the day, a dream team for any first-time director," he said.
Dhawan spoke about reuniting the stars after they last appeared together in comic release Garam Masala.
"Akshay was on board from day one, when I was still developing the story idea," he said. "A two-hero movie is never easy to write and it took me almost two years to put this project together.
"Once I felt I had justified the presence of both the protagonists, I took the script to John... he was convinced immediately."
After a number of cast changes, the London-based film will now feature Deepika Padukone and Chitrangada Singh.
Dhawan added that the project breaks the norms of conventional Bollywood comedy by not resorting to slapstick or exaggeration.
Desi Boyz is released by Eros International on November 25.
