The thriller, directed by AR Murugadoss, will be released in November to coincide with Diwali holiday season.
A source told Hindustan Times: "A number of Bollywood stars wanted to work with Murugadoss after Ghajini, but he was very impressed with Akshay's enthusiasm towards his work. The plan for this film was finalised early this year, but everything has been kept under wraps.
"Besides Thuppakki, Akshay will also star in the Hindi remake of Tamil hit Ramana, which will be produced by Sanjay Leela Bhansali Productions."
Murugadoss will be also be directing the remake of the film, which will be produced by Kumar and Vipul Shah.
Kumar's most recent hit Rowdy Rathore was also a Hindi language version of the Telegu film Vikramarkudu.
The action film - which also stars Sonakshi Sinha - has been the biggest success of Kumar's career so far.
