The story is set to reveal new secrets about the clone of Spider-Man, Ben Reilly, who died at the hands of Norman Osborn at the end of the Clone Saga in the 1990s.
Guggenheim told Marvel.com: "The original idea behind this story wasn't related to Ben or the Clone Saga at all.
"Rather, I was interested in the idea of a villain who hated Peter Parker, not Spider-Man. But Pete's such a nice guy. What could he do that would ever make someone mad enough to kill him?
"And then I tumbled to the idea that he wouldn't - but perhaps Ben might."
As well as exploring Reilly’s character, Guggenheim said that his swansong story features the return of fellow Spider-Man clone Kaine and villain Raptor.
Who Was Ben Reilly? begins in the pages of Amazing Spider-Man #608 - on shelves October 7.
