The marketing campaign features a logo for the Marvel miniseries designed by Rich Ginter emblazoned on a billboard at the Manhattan intersection.
Millar described the advertisement as "pretty much the most audacious marketing stunt in the history of comics".
Illustrated by Steve McNiven, Nemesis takes place outside of Marvel Universe continuity and features a character that has been described as a combination of DC's Batman and the Joker.
Previous marketing strategies for Millar comics have seen The Authority appear on the front page of The Times, a mock promotional video for Kick-Ass and the auctioning of character identities.
Nemesis #1 is available in stores now.
