Written by Johnston and illustrated by Sean Chen, the comic sees Matt Murdoch team up with fellow superhero Luke Cage.
"It's about friendship, and the respect you have to give someone to remain their friend," Johnston told CBR.
"It's about not backing down from impossible challenges, because to do so would jeopardise that respect. It's about Matt and Luke facing off in the abandoned, dusty ring at Fogwell's Gym for the ultimate match."
The scribe is also working on Marvel's ongoing Daredevil series, co-writing issue #506 with Andy Diggle. Cage Match marks his first solo Daredevil story.
Daredevil: Cage Match arrives in stores on May 12.
