The Penny Arcade creators appeared on the '2010 Time 100' list alongside Barack Obama, Oprah Winfrey and Lady GaGa.
The webcomic was founded in 1998 and draws an estimated 3.5 million viewers with its thrice-weekly posts.
With the aid of their business manager Robert Khoo, the pair went on to set up the Child's Play charity, which sends video games to sick children in more than 60 hospitals around the world.
In 2004, they also founded Seattle's Penny Arcade Expo, a convention dedicated to gaming culture.
Neil Gaiman and DC Entertainment president Diane Nelson were also in the running but did not make the final list.
