According to a statement from both firms, the new publication will dig into the "criminal past" of the Kane and Lynch characters, who first appeared in Square Enix's 2008 game Kane & Lynch: Dead Men.
The series has been written by Ian Edginton, with Chris Mitten and Ben Templesmith handling the artwork.
"We could not be happier about this partnership and expansion into comics. Who better to work with than DC Comics?" said IO Interactive general manager Niels Jørgensen.
"We hope the comic book world is ready for the violent mayhem of Kane & Lynch."
DC Comics' Hank Kanalz added: "Kane & Lynch are two of the most interesting video game characters I have ever seen.
"I think this new series will show the diversity of our line of books, as our creators capture the essence of these two criminals."
The Kane & Lynch comic book will be produced by DC's WildStorm imprint, with the first issue scheduled for launch in August.
New game Kane & Lynch 2: Dog Days will be released on PS3, Xbox 360 and PC on August 27.
