Dating back to 1986, the artwork originally appeared on issue #313 of the comic and depicts the superhero battling the Lizard.
According to Bleeding Cool, the illustration is not one of the artist's high-profile covers. It is thought that his designs for Amazing Spider-Man #300 or #301 could fetch significantly higher prices.
The website speculated that the item was purchased by Wizard Entertainment CEO Gareb Shamus, who owns an extensive collection of Amazing Spider-Man cover art by McFarlane.
Earlier this year, a Daredevil cover illustration by Frank Miller made over $100,000 (£70,000) when it went under the hammer.
