Dwayne McDuffie, a comic book writer well known for his work in producing and writing the animated series Justice League Unlimited, will script the adaptation.
Comic Vine reports that Desperate Housewives star James Denton will take on the lead voice role of Clark Kent/Superman, while Mad Men actress Christina Hendricks will play Lois Lane.
Denton becomes the latest in a number of actors to take on the iconic role in recent years, with Adam Baldwin voicing Superman in Superman/Doomsday, and another Desperate Housewives star Kyle MacLachlan voicing him in New Frontier.
Rounding out the cast are Anthony Lapaglia as Lex Luthor, Ed Asner as Daily Planet editor Perry White, and Six Feet Under alumna Frances Conroy as Ma Kent.
All-Star Superman is due to be released early 2011.
