According to writer and artist Nigel Parkinson, the strip is based on a concept by the TV Burp host, who also contributed jokes and oversaw its development.
Hill's writing team - led by David Quantick - will script the short in future issues.
"The Dandy has had an injection of fresh talent and ideas, yet maintains its 70 plus years tradition of knockabout humour," Parkinson told DS. "I was happy to be able to bring Harry Hill along to join the fun."
Publisher DC Thomson has also confirmed that Top Gear host Jeremy Clarkson will appear on the cover of a future issue.
The Dandy relaunches on October 27.
