The pair will collaborate on a three-issue arc beginning in Supergirl #65 in June, reports Robot 6.
It will mark DeConnick's debut with DC Comics. She previously co-wrote 30 Days of Night: Eben & Stella with Steve Niles and wrote Marvel's Osborn miniseries and Sif and Rescue one-shots.
ChrisCross has drawn various titles for DC including Firestorm, The Outsiders and Superman/Batman.
Supergirl has been subject to repeated creative team changes since the departure of regulars Sterling Gates and Jamal Ingle from the title in December.
