Titled 50 Cent: Blood On The Sand, the game follows the million-selling but critically-panned 50 Cent: Bulletproof, which was released in 2005.
The sequel was previously dropped by Activision Blizzard, but has now been snapped up by THQ.
Brad Carraway, THQ's vice president of global brand management, described the rapper as a "cultural icon" and said: "THQ is thrilled to be working with him on this exciting new title."
Carraway indicated that Blood On The Sand will combine single play as well as "drop-in and drop-out online co-op". It will also feature 18 previously unreleased tracks from 50 Cent.
The plots focuses on the rapper and his G-Unit fighting their way across war-torn nations in order to collect on a debt owed by the boss of a crime cartel.
50 Cent: Blood On The Sand is reportedly scheduled for release on Xbox 360 and PS3 in the first quarter of 2009.
