Tracks by The White Stripes, Bob Dylan and Johnny Cash will appear on the title, while players will be able to play through the songs with any combination of the four instruments available.
In addition to custom band play, which could see the use of up to four guitarists or two drummers and two singers, the game will also feature a party mode, enabling players to drop in and out of songs at will.
Activision also revealed that the latest title would contain five battle modes, as well as a challenge mode that requires songs to be played in a specific way.
Guitar Hero 5 is due for release this autumn.
The current set list:
The Rolling Stones – 'Sympathy For The Devil'
Bob Dylan – 'All Along The Watchtower'
