According to Blabbermouth.net, The Rocky Horror Picture Show star will take on the role previously assigned to heavy metal singer Ronnie James Dio.
No specific reason is currently known for the change.
It was confirmed earlier this month that Black Sabbath singer Ozzy Osbourne will also feature in the game as the Guardian of Metal.
Jack Black has leant his voice and likeness for the lead role of roadie Eddie Riggs, while Judas Priest's Rob Halford and Motorhead frontman Lemmy will also make appearances.
Brütal Legend will be released in Europe for Xbox 360 and PS3 on October 16.
