The Grammy Award-winning hip-hop artist - real name Jeffrey Townes - will join DJ Shadow, DJ Z-Trip, Grandmaster Flash and DJ AM as characters in the music simulation title.
He will also provide two exclusive mixes entitled LL Cool J's 'Rock the Bells' Vs The Aranbee Pop Symphony Orchestra doing 'Bittersweet Symphony' and Tears For Fears's 'Shout' Vs Eric B. & Rakim's 'Eric B. is President'.
"I am very, very excited DJ culture is finally getting its due. To have a song in DJ Hero is a dream come true; to be a character someone can pick up the controller and select you and play is probably the biggest thing to happen to me in my career," said Townes.
"Music is so much a part of everyone's life, DJ Hero is going to appeal to almost everybody."
In development by FreeStyleGames, DJ Hero is to feature 100 songs and 80 unique mixes of music ranging from rap to electronica to R&B.
It will be released on Xbox 360, PS3 and Wii in November this year, alongside a turntable-style controller for scratching and mixing.
