Gary Oldman will reprise his role of Vicktor Reznov from World At War for the Cold War-era shooter.
He will be joined by Apollo 13 and The Rock actor Ed Harris, who will voice CIA operative Jason Hudson.
It was also revealed that Dark Knight and Superman: Man Of Steel scriptwriter David S. Goyer has been a consultant for the game's story.
"Oldman, Harris and Goyer are considered some of Hollywood’s finest talents and they perfectly complement Call of Duty: Black Ops' ambitious and immersive single-player experience," said Treyarch studio head Mark Lamia.
"Their contributions have helped us to push the boundaries of our story telling and character development far beyond anything we have ever attempted before in the franchise."
Black Ops will be available for Xbox 360, PS3 and PC from November 10.
