The firm has confirmed that 482 GAME and Gamestation stores will open at midnight tonight to sell the device.
Each store will also be giving away a Nintendo 3DS free to one customer, with attendees receiving a raffle ticket for a prize draw on the night.
GAME's Oxford Street branch will host a special launch event featuring comedian Iain Lee, and will hand out a free 3DS game to the first 100 people queuing outside.
The highstreet chain will also be hosting five of the 11 VIP launches across the country, including at its stores in Birmingham, Cardiff and Manchester.
"Thursday night is a must-attend moment for Nintendo fans - the 3DS is a stunning piece of gaming tech which impresses everyone who plays it," said head of PR Neil Ashurst.
"We are opening 482 of our stores at midnight to allow our customers to be the first to get their hands on it. Whatever store they go to, we will be offering a free 3DS console for one lucky individual, so it's definitely worth turning up for midnight!"
The firm will also offer a unique trade-in offer for those who return the device within the first month of purchase.
The Nintendo 3DS will be available across the UK from midnight tonight, and will debut alongside 15 launch titles.
