The puzzle adventure title will feature over 270 mini-games and puzzles from a variety of genres, and has been teased to last 25 hours.
"Puzzle adventures, hidden object games and rhythm action titles make for some of the most popular handheld games," said product director Neil McKenna .
"But never before has a game offered all three - and so much more. May's Mysteries: The Secret of Dragonville is an incredible piece of software which will captivate all who play."
He added: "We think May could become the poster child for puzzle adventures, and we can't wait for DS owners to fall in love with a new heroine this August."
May's Mysteries will be the latest puzzle adventure title for the DS. Nintendo releases Professor Layton annually, while the BBC launched Doctor Who: Evacuation Earth last year.
Elsewhere, Konami's Doctor Lautrec and the Forgotten Knights will be available on Nintendo 3DS this summer in Japan.
May's Mysteries: The Secret of Dragonville is developed by V5 Play and will be available in Europe this August.
