The Deus Ex creator will pick up the prize at the Game Developers Choice Awards, held on March 7.
Spector is also hailed as the mastermind behind Thief and System Shock. He is currently heading up the Disney-owned developer Junction Point Studios.
GDC general manager Meggan Scavio said Spector's career to date "exemplifies the exact qualities that a 'Lifetime Achievement Award' recipient should possess."
Previous winners of the GDC Lifetime Achievement Award include Nintendo's Shigeru Miyamoto, Metal Gear Solid designer Hideo Kojima and Civilization creator Sid Meier. Lionhead Studios leader Peter Molyneux picked up last year's prize.
The Game Developers Conference 2012 takes place between March 5 and March 9.
