The British actor and comedian confirmed the role on Twitter, though it remains unclear if it is for Assassin's Creed 3 or PlayStation Vita title Liberation.
"Did a voiceover for the new Assassin's Creed (looks great btw) - got to say "PEGI 18". Always makes me think of @simonpegg," he tweeted.
The Shaun of the Dead and Spaced actor has previously recorded lines for 2003 release Warhammer 40,000: Fire Warrior.
He also reprised his role as the voice of Darth Maul in Star Wars: Battlefront II in 2005.
Announced in March, Assassin's Creed 3 will take place over a 30-year period in colonial America beginning in 1777.
It features a brand new lead named Connor, the offspring of an English father and Native American mother.
The title is on course to break Ubisoft's record for the most pre-ordered game of all time.
Assassin's Creed: Liberation, meanwhile, was announced at last week's E3 Gaming Expo.
It will feature the series' first ever female protagonist, and support cross-play with the PlayStation 3 edition of Assassin's Creed 3.
Both versions are scheduled for release on October 30.
