Titled The Last Sermon, the latest video introduces the Horseman Death, who faces off against legions of beasts.
The dramatic live action trailer begins in a church besieged by demons, before help arrives in an unexpected form.
An extended version can be viewed on the game's Facebook page, while a TV spot will debut during tonight's (June 19) episode of Leverage at 10pm on FX.
The story of Darksiders 2 will run parallel to that of the first Darksiders game. Players will take on the role of the horseman Death as he travels through the Abyss to redeem his brother War and undo the apocalypse.
THQ previously announced a number of pre-order bonuses for Darksiders 2 available at various retailers.
The game was originally due to launch on June 29, but was recently pushed back until an unspecified point in August.
The delay is said to have nothing to do with the recent spate of job cuts at Vigil Games.
Darksiders 2 will release for the PS3, Xbox 360 and PC. A Wii U version is also said to be in development.
