The cinematic trailer, which was unveiled at this year's Comic-Con event, introduces a brand new character named Bill.
Taking place at Bill's safe house, the video begins with the trio escaping a group of Infected, before planning their next move.
It was also revealed that Bill would be voiced by Hollywood actor W. Earl Brown, who has appeared in movies such as There's Something About Mary, Scream and Vanilla Sky, as well as TV shows including Deadwood.
Announced at last year's Spike TV VGAs, The Last of Us follows two survivors in a world turned upside down by a fungus-like virus.
The adventure will be set in a more colourful world than other post-apocalyptic games, using overgrown vegetation in cities to add variety.
Both gunplay and melee combat will play an important part in the game, with weapons breaking after continued use, requiring the player to find new items regularly.
Work on the project began after Uncharted 2: Among Thieves and it has been in development for over two years.
The development team recently revealed that the game would not support co-operative multiplayer.
The Last of Us will be released exclusively on PlayStation 3 in 2013.
