The rapper and media mogul was unveiled as executive producer yesterday.
He will focus his efforts on the game's presentation, hand picking the soundtrack, designing the interactive menus and contributing to the "look and feel" of the title.
"This has been a unique opportunity to collaborate with 2K Sports and be a part of one of the best sports video games in history," revealed Jay-Z.
"NBA 2K13 will be the next evolution in sports and basketball culture, and I'm ready to usher in the new era of the franchise," he added.
"Jay-Z's role in NBA 2K13 represents a unique fusion of basketball, music, art, and entertainment, and illustrates the evolution of NBA 2K into something much bigger than simply the best basketball video game series ever made."
Yesterday's debut NBA 2K13 video features the likes of Michael Jordan, Blake Griffin, Kevin Durant and Derrick Rose.
The game releases for the Xbox 360, PS3, PlayStation Vita, Wii and PC on October 2 in North America. A European launch date is yet to be finalised.
It will also release for the Wii U. Listed for a "holiday" launch, it will likely be available within the first few months of the console's release.
Previous NBA 2K games have been well received, earning near-unanimous praise for their presentation, depth and polish.
