The latest gameplay trailer details the game's multipalyer, which includes deathmatch, competitive and co-operative modes.
In addition to the standard deathmatch mode, players will be able to join the co-operative Wolf Pack mode where two players must work together to eliminate groups of targets in the allotted time.
There will also be the team-based Domination mode, where teams compete to control territory across the map in order to win.
The multiplayer maps will take players to locations outside of the single-player campaign and with a variety of weather effects, ranging from Boston Harbor to a Virginia plantation.
There will be 16 character classes for multiplayer, each with their own unique appearance, weapons and skills. Players will also be able to choose from 14 abilities and 10 character perks to further customise their play style before each match.
Officially unveiled in March, Assassin's Creed 3 will take place over a 30-year period in colonial America beginning in 1777.
It will be accompanied by PlayStation Vita spinoff Assassin's Creed 3: Liberation, which was announced at this year's E3 Gaming Expo.
Liberation will support cross-play with the PlayStation 3 edition of Assassin's Creed 3, allowing players to unlock bonuses in both games by trading goods.
Ubisoft recently confirmed that it will offer a season pass for Assassin's Creed 3 DLC, which will include new multiplayer maps and the 'Tyranny of King Washington' single-player expansion.
Assassin's Creed 3 and Liberation will be released for Xbox 360, PlayStation 3 and PlayStation Vita on October 31 worldwide.
A Wii U version is also in development and will be available during the system's launch window this holiday season.
