Developed by Traveller's Tales, the game will be available for the Xbox 360, PS3, PC, Wii U, 3DS, DS and Vita this autumn.
Featuring more than 100 Marvel characters including Hulk, Thor, Spider-Man, Wolverine, Captain America and Black Widow, the game sees players attempt to save the world from the threat of Loki and Galactus.
Characters will have individual abilities, which will be used to solve puzzles and battle enemies. It will also contain the series's usual dose of humour.
Hinting at some of the game's environments, Traveller's Tales managing director Tom Stone said: "We're very excited to bring a myriad of classic Marvel characters to life in LEGO form for some fun, family-friendly gameplay in LEGO Marvel Super Heroes.
"Players will brave the ultimate mission to save the world as they explore famous landmarks in LEGO New York and take the adventure beyond Earth to iconic locations in the Marvel Universe."
Marvel's TQ Jefferson, meanwhile, described LEGO Marvel Super Heroes as the game fans had been craving for years.
"Fans of both LEGO and Marvel have wanted this game for years, and we're addressing that demand with more Marvel Universe characters than ever in a console game," explained Jefferson.
"If you're a fan of Spider-Man, the Avengers, Fantastic Four, X-Men, Guardians of the Galaxy or scores of other Marvel characters, this is the game for you," he added.
The game will complement LEGO Marvel construction sets based on Spider-Man and X-Men, as well as imminent sets taken from Iron Man 3.
Traveller's Tales has also released a number of games based in the DC universe, including last year's LEGO Batman 2: DC Super Heroes.
Other LEGO games include LEGO Lord of the Rings, LEGO Harry Potter and upcoming Nintendo exclusive LEGO City Undercover.
