Kay will front the show from March 2 to March 9 together with Moyles's usual team, Music Week reports.
He said: "I am looking forward to stepping into the big man's shoes.
"We'll be getting right behind Chris and the team as they climb Kili and bringing listeners the latest news from up the mountain."
Moyles used his Twitter page to confirm Kay's appointment for the six-day stint.
He will be joined by Gary Barlow, Alesha Dixon, Ben Shephard, Denise Van Outen, Fearne Cotton, Ronan Keating, Cheryl Cole and Kimberly Walsh on the climb.
