The DJ, 25, joined the station in February 2003 as a replacement for long-serving chart host Mark Goodier. He will front his final show next February, almost two years to the date.
Replacing Butters are newcomers JK & Joel, who will take the helm of a "brand new" chart show featuring download and album charts alongside the traditional singles rundown.
Ben Cooper, head of mainstream programming at Radio 1, said that the pair would "breathe new life" into the station's biggest show.
Also in February, Sara Cox will make her return to the station after eight months of maternity leave with a new weekend afternoon show. Scott Mills will remain as host of weekday drivetime.
"I'm looking forward to being back, I've really missed being on the radio," Cox said today. "I'm going to do my best not to burst into nursery rhymes although The Wheels On The Bus is an old school classic."
R1 controller Andy Parfitt added: "We are beginning to reap the benefits of the changes we initiated towards the end of last year. These latest moves are simply part of the continuing evolution of the station."
