The broadcaster joined the station in 1993 and fronted The Evening Session with Jo Whiley and later by himself, as well as hosting Lamacq Live and more recently In New Music We Trust.
Lamacq, who will continue working on Radio 2 and BBC 6 Music, said: "It was a wonderful opportunity to try and subvert pop music radio from within, which I hope we managed to do more often than not."
Monday nights will now feature music documentaries Radio 1 Stories from 9 to 10pm and Wednesdays will see Huw Stephens present programmes on new musical experimentation.
Tim Westwood's Tuesday night show will be replaced by a discussion programme hosted by Nihal, while the length of Westwood's Saturday night show will be halved to two hours before Mistajam takes over.
Thursdays will see newer names on Radio 1 with the 9 to 10pm slot of In New DJs We Trust.
Pete Tong and Annie Mac swap their Friday night dance slots, with Tong taking the 9 to 11pm session and Mac DJing from 7 to 9pm.
Deputy controller of Radio 1 Ben Cooper said: "Radio 1 must continue to evolve to help sharpen our offer to younger audiences.
"We're committed to delivering the best content to our audiences and our refreshed schedule reflects this."
It was recently announced that Fearne Cotton would replace Jo Whiley on the station in her weekday morning slots.
