On February 1, the renowned author will take charge of the 34th keynote lecture in honour of veteran broadcaster Dimbleby, who passed away in 1965.
Since his first Discworld novel The Colour Of Magic was published in 1983, Pratchett's acclaimed series has sold over 65 million copies around the world and been translated into 37 languages.
However, the writer has also recently discussed his battle with a rare form of early-onset Alzheimer's disease.
Titled Shaking Hands With Death, Pratchett's keynote lecture will explore the ways in which modern society is coping with its ageing population, particularly in terms of those people suffering with incurable illnesses.
The author, who was knighted last year, will discuss how death must be redefined in this new world. The lecture will be produced by Victoria Simpson and executive produced by Phil Dolling.
"I'm absolutely delighted that one of our most popular and best-loved authors has agreed to give this lecture," said controller of BBC One Jay Hunt.
"Sir Terry Pratchett has spoken with great bravery and honesty about his battle with Alzheimer's and I look forward to an intelligent and thought-provoking speech."
The Prince of Wales, Dame Stella Rimmington, former US President Bill Clinton and Dr Rowan Williams have all previously given Dimbleby lectures.
