According to The Guardian, MacIntyre - who joined London Tonight in April - will co-present his final edition of the show on September 17.
Despite leaving the programme, MacIntyre will continue to work with London Tonight's producer ITN on a freelance basis as a journalist and presenter.
ITN said that it is now searching for a permanent co-anchor to join Nina Hossain on the show.
MacIntyre, who appeared on ITV's Dancing On Ice last year, is currently working on a number of projects, including a development deal in the US.
"I've had an absolutely fantastic time working at London Tonight and feel privileged to continue to work within ITN on a freelance basis," he said.
"My American project and my directing opportunities are simply too demanding for me to devote enough time to London Tonight."
ITV's London Tonight editor Faye Nickolds added: "Donal has done a great job at London Tonight, and his exciting opportunity in the US means that he'll be best working with us in a freelance capacity.
"We wish him all the best with his future work and look forward to him continuing to contribute to our programme. We'll be announcing a permanent co-presenter with Nina Hossain in the near future."
