The Sunrise host had initially seemed surprised when questioned by one of his Twitter followers, asking where they had heard the news. However, Holmes soon clarified that he will front the broadcast on April 29.
"Yes it is true... Just wasn't public," he wrote. "It's a big thing and a great honour to be given the responsibility by Sky News."
The Mirror had reported the story yesterday, adding that Holmes will be supported by other presenters including Kay Burley and royal correspondent Sarah Hughes.
The BBC has confirmed that Huw Edwards will lead its coverage of the event. ITV's royal wedding coverage will be fronted by This Morning host Phillip Schofield and News at Ten anchor Julie Etchingham.
Holmes picked up the 'Satellite/Digital TV Personality' prize at the TRIC Awards 2011 earlier this month.
