Mykura will replace Peter Dale, the digital-only channel's launch head who announced his departure last week.
Channel 4 director of television Kevin Lygo, to whom Mykura will report, said: "Through its close association with the British Documentary Film Foundation, More4 champions original documentaries from around the world, and so I am delighted that Hamish will be bringing his expertise in this area to the channel."
Mykura's credits include Diana: The Witnesses In The Tunnel and Enemies Of Reason, the Richard Dawkins miniseries.
He was made head of documentaries earlier this year and, according to Channel 4, has seen the audience for its headline Cutting Edge strand increase by 57% year-on-year.
Mykura has previously worked at Blakeway Productions, Mentorn and for ten years at the BBC.
