The former couple, who first met on the set of 1999 movie Pushing Tin, have managed to remain friends following the break-up of their three-year marriage in 2003.
Thornton has now revealed that Jolie is keen to appear on the big screen with him again - but they would not feel comfortable playing a married couple.
"We've talked about it plenty of times," Thornton told Maxim magazine. "I'm sure we will."
He continued: "We want to be really careful that we pick the right one. Maybe a comedy. In other words, we wouldn't want to do another movie about a husband-wife relationship. That probably wouldn't be very good."
Jolie and her partner Brad Pitt welcomed the arrival of twin children in France earlier this month.
