According to This Is Plymouth, Disney has pencilled in the Lewis Carroll adaptation for a two-week shooting stint in the coastal city.
Extras are currently being sought for the film, which begins production next month.
Casting agent Ilenka Jelowicki said: "We are looking for people above the age of 16 who have naturally coloured hair.
"We would prefer girls with long hair and men with natural styles. Extras will get to dress up in Victorian period costume, corsets and wigs and will have their makeup done."
Locations have yet to be confirmed, although Plymouth's Saltram House, Mount Edgcumbe and the Royal William Yard are thought to be strong possibilities to feature in the film.
Australian actress Mia Wasikowska has been cast in the lead role, with Johnny Depp rumoured to be playing The Mad Hatter.
