The duo, who are writers and producers on the American version of The Office, will work on the first big screen instalment in the franchise for 19 years.
It is not known whether the original cast of Ernie Hudson, Dan Akroyd, Harold Ramis and Bill Murray will be returning for the project.
However, Stupnitsky and Eisenberg recently worked with Ramis on his biblical comedy Year One, which is released next year.
Original director Ivan Reitman is involved in the movie's development, according to The Hollywood Reporter.
The original Ghostbusters, which was released in 1984, grossed $292 million at the worldwide box office. A second movie was released in 1989.
In July it was reported that Steve Carrell and Seth Rogen were being lined up for a sequel to the ghost-hunting adventures.
