Gugino will play a co-worker of Schreiber's TV writer who pushes his marriage to Hunt's character to breaking point, reports Variety.
Directing debutant Levine will helm from his own script, with production commencing in New York this month.
Gugino can currently be seen opposite Robert De Niro and Al Pacino in Righteous Kill. She recently completed filming a role in Disney's Race To Witch Mountain.
