Developer Electronic Arts (EA) has finalised a deal with Universal Pictures to bring the story, a tale of two Army soldiers turned mercenaries, to the big screen.
Game sales have grossed more than $100 million (£63m) since its release in March 2008, according to EA.
Production of the film will begin in 2009, with Role Model's Scott Stuber overseeing and Bourne Ultimatum screenwriter Scott Z Burns handling the script.
"Army of Two is a great project that has already attracted top name talent, and we're glad to be working with Electronic Arts on making this hit game into a film franchise," said Universal Pictures president Donna Langley.
