The Universal drama follows Vaughn's character as he returns home to New York with massive gambling debts. There he finds his mother is dying of cancer and is setting up his old girlfriend to look after a 5-year-old girl, who is actually his daughter, after she passes away.
O'Connor, whose new film Pride And Glory is about to hit UK cinemas, will direct Sunny And 68 from a script he co-wrote with Anthony Tambakis.
"It's a story of how three generations of women teach this character to become a man," O'Connor told Variety.
