Vince Vaughn, Kristen Bell, Jason Bateman, Kristin Davis and Jean Reno are also confirmed for the movie.
Serafinowicz will play British hotelier Stanley, who owns the Eden West Resort that the couples reside in.
The comic's credits include The Peter Serafinowicz Show, I'm Alan Partridge, Spaced and Shaun Of The Dead.
Shooting is currently taking place in Bora Bora.
