Kendrick, who has a small role in the forthcoming Twilight, was chosen above better-known actresses such as Ellen Page and Emily Blunt for the sought-after role.
She will play a young woman who is pulled into the life of Clooney's character Ryan Bingham, a businessman who is obsessed with collecting one million frequent flier miles.
Reitman, who previously directed Juno, wrote Up's screenplay based on Walter Kirn's satirical novel.
