Winslet picked up two awards, winning best drama actress for Revolutionary Road and best supporting actress for The Reader.
Accepting her second prize for Revolutionary Road, she apologised to her fellow nominees, saying: "I'm so sorry … Is this really happening?" before paying tribute to her "spectacular" co-star Leonardo DiCaprio and husband Sam Mendes, who directed the film.
Ledger was given a posthumous supporting actor award for his role as the Joker in The Dark Knight. The film's director Christopher Nolan accepted the honour on his behalf, saying: "All of us who worked with Heath accept with an awful mixture of sadness but incredible pride."
He added that Ledger would be "eternally missed but never forgotten".
However, the biggest winner of the night was Danny Boyle's Slumdog Millionaire, which picked up all four awards that it was nominated for, including best film drama and best director.
Other winners included British actress Sally Hawkins who was named best comedy actress for her role in Happy-Go-Lucky and Colin Farrell who scooped best comedy actor for In Bruges.
Mickey Rourke picked up best drama actor for The Wrestler, while Woody Allen's Vicky Cristina Barcelona was named best film comedy.
Steven Spielberg was awarded the Cecil B. DeMille award for lifetime achievement.
