Dushku had purchased the rights to co-produce the movie and has already cast her brother Nate in the lead role.
The film, entitled The Perfect Moment, will be the first feature directed by Timoner, who is best known for her Brian Jonestown Massacre documentary DIG!, Variety reports.
Timoner said: "A pariah to some, a hero to others, Mapplethorpe's talent combined with his irreverence toward social norms made him a cultural lightning rod.
"He is an artist who could not help but express his vision of the world, despite the tempest it spawned."
The Mapplethorpe Foundation said that it was "impressed" with Timoner's vision for the project.
