The project, which was announced last year, will see original Blue Men Matt Goldman, Chris Wink and Phil Stanton return to star as the mute performers.
Describing the storyline to Variety, Russo said that it will involve the trio "entering the brain of a socially and creatively congested person and observing his neural patterns and his habitual brain functions and memory and altering it in a way that helps him bring his inside life outside".
Wink added: "We want all our work to touch you in a simple way. We're vaudevillians; we're not here with deep messages."
Charlotte Huggins (Journey To The Centre Of The Earth) is producing the movie, which is due to open on IMAX screens in early 2011.
