The family comedy, a combination of live-action and CG animation, will revolve around an unemployed man who runs over the Easter Bunny, prompting him to rescue the holidays when the injured animal is unable to walk.
Alvin and the Chipmunks's Tim Hill is directing I Hop, with Chris Meledandri and John Cohen producing.
Speaking to Variety about Brand's casting, Meledandri said: "Russell showed me that he's got a wonderful ability not only to be funny in his own body, but he can create humour vocally, which is the distinction we need for these movies.
"This gives us an opportunity to remythologise the holiday around an Easter Bunny character that is as dynamic and irreverent as Russell is."
I Hop has been pencilled in for a March 4, 2011 release.
