Speaking to MTV at Comic-Con, the Lost star claimed that he is in the first round of discussions about playing the superhero.
Akinnuoye-Agbaje said that his role in forthcoming blockbuster GI Joe may help him land the role.
"Hopefully they'll look at GI Joe and see the potential," he commented. "But it's about time we have a black superhero, isn't it?"
He added: "[The Black Panther is] from a fictional village in Africa and the timing is so right for that kind of character to come through.
"And while I'm in my prime, this is the time. We've got [Barack] Obama, now we need something on screen to represent."
Akinnuoye-Agbaje can be seen in GI Joe: The Rise Of Cobra from August 7.
